gene expression profiling for cancer targeted therapy

چکیده
background : lung cancer is still the leading cause of cancer- related deaths and most of themâ are normally diagnosed in late stages, so there is an urgent need to establish reliable tools for the identification at early stages of the disease. in this study, we use an eleven gene signature test to examine lung biopsy specimens for diagnostic, prognostic & therapeutic purposes. methods : bronchoscopy was performed to collect 102 fresh frozen specimens from 51 patients. 32 lung tumors & their adjacent normal tissues were examined along with biopsy specimens. in all specimens, total rnas were isolated & after generating cdna, were used for qrt-pcr analysis. results : the results indicate that the expressions of 3 oncogenes bcl-2, k-ras & h-ras were significantly high (p<0.05) in surgically removed tumors as well as biopsy specimens compare to adjacent normal tissues. the differential expressions of these 3 genes were strikingly higher in cigarette using patients than nonsmokers. conclusion: considering the overall results of this study, it can be concluded that, lung tissue abnormalities other than cancer could cause the elevation of some oncogene expression. this study also verifies that smoking can definitely cause oncogenic mutation, activation &/or amplification; which is now a globally proven fact.

Targeted cancer therapy: review article
Cancer is one of the most dangerous health problems of today modern societies which has an increasing rate especially in developing countries. There are many diverse ongoing treatment attempts trying to defeat cancer. Despite that, scientists have been unable to find a permanent cure for this disease. In many cases although there is a successful first response in patients, cancer cells are fina...
متن کاملCancer Gene Therapy: Targeted Genomedicines
Given the fact that malignant cells proliferate more rapidly than normal cells, damage to the cancer cells is anticipated to be markedly greater than normal cells. However, cancer cells generate chemoresistance mechanisms within the tumor microenvironment, while undesired toxicity may occur in the normal cells. For example, in colorectal cancer (CRC), there exist well-described sequences of mut...
متن کاملTargeted, non-viral gene delivery for cancer gene therapy.
The ability to mediate targeted and specific delivery of therapeutics to cancer cells remains one of the most important hurdles in effectively treating cancer. This aspect also remains as one of the greatest limitations of gene therapy as well. Targeted vectors based on the use of DNA-binding agents attached to cell specific ligands or "molecular conjugates" were created with the goal of over-c...
متن کاملTargeted Expression of Suicide Gene by Tissue-Specific Promoter and MicroRNA Regulation for Cancer Gene Therapy
In order to realise the full potential of cancer suicide gene therapy that allows the precise expression of suicide gene in cancer cells, we used a tissue specific Epithelial cell adhesion molecule (EpCAM) promoter (EGP-2) that directs transgene Herpes simplex virus-thymidine kinase (HSV-TK) expression preferentially in EpCAM over expressing cancer cells. EpCAM levels are considerably higher in...
متن کاملGene expression profiling reveals U1 snRNA regulates cancer gene expression
U1 small nuclear RNA (U1 snRNA), as one of the most abundant ncRNAs in human cells, plays an important role in splicing of pre-mRNAs. Compared to previous studies which have focused on the primary function of U1 snRNA and the neurodegenerative diseases caused by abnormalities of U1 snRNA, this study is to investigate how U1 snRNA over-expression affects the expression of mammal genes on a genom...
متن کاملTargeted Therapy for Cancer
Cancer is one of the most common causes of death, taking nearly 7 million lives each year worldwide. New cancer targeted therapies that make use therapeutic antibodies or small molecules have made treatment more tumor specific and less toxic. Nevertheless, there remain several challenges to the treatment of cancer, including drug resistance, cancer stem cells, and high tumor interstitial fluid ...
